一、多能干细胞状态的转换和体细胞重编程的分子机制
1. 聚焦重要的转录因子SALL家族在多能干细胞干性维持、状态转换和三胚层分化过程中的重要作用和分子机制。
2. 运用CRISPR/Cas系统筛选与体细胞重编程和多能干细胞分化相关的新基因并研究其机制。
二、基于多能干细胞的心脏生物起搏和再生治疗,探究调控心脏发育的分子机制,以阐明心脏疾病的发病机制及新的治疗靶点的研发
1. 通过建立高效的心脏起搏细胞分化方法获得功能性的起搏细胞为生物起搏器的研发提供细胞来源。
2. 通过诱导多能干细胞分化为心房肌细胞,在体外建立房颤的模型(2D, 3D),探索应用新型纳米材料进行房颤的实验性治疗。
3. 研究参与早期心脏发育的重要转录因子的调控机制,通过干细胞来源的心肌细胞建立心脏疾病的体外模型,研发干预的新靶点。
